Method of Production :

There are many methods for preparing R-lipoic acid through chemical synthesis in the world, and there are mainly three types of methods:
1. Starting from 6,8-dichlorooctanoic acid ethyl ester, racemic α - lipoic acid was obtained through thiolation, cyclization, and hydrolysis (U.S. 27924061957); U.S.3223712,1965), The separation agent was used to obtain R-lipoic acid, and the yield of this step was only 45%. And S-lipoic acid is not easily converted to R S-type or R-type waste half of resources and make R-lipoic acid expensive.
2. Using 6,8-dihydroxyoctanoic acid methyl ester or 6-hydroxy-8-chlorooctanoic acid methyl ester as the starting material, methanesulfonate is prepared, and then stereoselective to R-lipoic acid (U.S. 5869713), G.Bringmann, J.Paust Z.Naturforschung 54b,661-665,1999), But the process is relatively complex and it is not easy to obtain pure products.
3. Hydrolyze racemic 6,8-dichlorooctanoic acid ethyl ester into (±) dichlorooctanoic acid (DCA), salt with (-) ephedrine, split, thiolate, cyclize (Acker, etc.), J.Am.Chem.Soc., 76,6483,1954), This method can basically save costs, but about 50% of S - (-) -6,8-dichlorooctanoic acid is still not utilized, resulting in higher costs.
There are no detailed literature reports on the conversion of S - (-) -6,8-dichlorooctanoic acid to R - (+) -6,8-mercaptooctanoic acid and R-thiooctanoic acid.
